چکیده

Part of this work was previously reported at the symposium "Diabetes Research Today," held in Capri, Italy, 13-15 April 1976, and published in Symposia Medica Hoechst 12, 223240 (1976), Friedrich-Karl Schattauer-Verlag GmbH, Stuttgart, West Germany. Dr. Cerasi's present address is the Department of Endocrinology and Metabolism, the Hebrew University Hadassah Medical School, Jerusalem, Israel. Received for publication 25 July 1977 and in revised form 8 November 1977. hibited both the glucose and the sulfonylurea [3H]cyclic AMP and insulin responses. Epinephrine (1 uM) partially inhibited the [3H]cyclic AMP response to both glucose and sulfonylurea, whereas insulin release was completely abolished. It is concluded that the sulfonylurea effects on islet cyclic AMP are intimately related to those of glucose. It is suggested that sulfonylureas exert a major part of their action by facilitating the effect of glucose on the beta-cell adenylate cyclase; the increased cyclic AMP level, in its turn, enhances the secretion rate of insulin.

Effects of Sulfonylureas on Peroxisome Proliferator-Activated Receptor γ Activity and on Glucose Uptake by Thiazolidinediones

BACKGROUND Sulfonylurea primarily stimulates insulin secretion by binding to its receptor on the pancreatic β-cells. Recent studies have suggested that sulfonylureas induce insulin sensitivity through peroxisome proliferator-activated receptor γ (PPARγ), one of the nuclear receptors. In this study, we investigated the effects of sulfonylurea on PPARγ transcriptional activity and on the glucose ...

Epac2: a sulfonylurea receptor?

Sulfonylureas are widely used oral drugs in the treatment of diabetes mellitus. They function by the inhibition of ATP-sensitive K+ channels in pancreatic β-cells, which are thus considered the 'classical' sulfonylurea receptor. Next to the ATP-sensitive K+ channels, additional sulfonylurea-interacting proteins were identified, which might contribute to the physiological effects of this drug fa...

Effect of Sulfonylureas Administered Centrally on the Blood Glucose Level in Immobilization Stress Model

Sulfonylureas are widely used as an antidiabetic drug. In the present study, the effects of sulfonylurea administered supraspinally on immobilization stress-induced blood glucose level were studied in ICR mice. Mice were once enforced into immobilization stress for 30 min and returned to the cage. The blood glucose level was measured 30, 60, and 120 min after immobilization stress initiation. W...

Recurrent and persistent hypoglycemia following intoxication with glibenclamide overdose: case report

Background: Hypoglycemia can cause permanent damage to the brain or lead to death. That is why it is very important to prevent or quickly correct hypoglycemia to save life for the treatment of hypoglycemia following the use of Sulfonylureas, taking oral nutrition for the patient or administration of injectable hypertonic glucose is recommanded. But recurrence and resistance to treatment are com...

EVALU ATION OF INSULIN-SUL FONYLUREA COMBINATION V ERSIJS INSULIN AL ONE IN THE T REATMENT OF NON-INSULIN D EPENDENT DIABETES MELLITU S (NID DM)

Lowering the glucose level in diabetic patients reduces the risk of many consequences which are usually reported in these patients. Non-insulin dependent (type II) diabetes which cannot be controlled with sulfonylureas alone can be controlled with a combination of sulfonylurea and insulin. In an open randomized trial 58 patients were divided into three groups. The fIrst group received only ...
